      以玉米秸秆纤维素乙醇残渣、芦苇、核桃壳和稻壳4种农业废弃物为原料,采用氢氧化钠法分别提取木质素。通过凝胶渗透色谱、傅里叶变换红外光谱、核磁共振等分析手段对获得的木质素进行结构分析,探究了不同原料来源木质素的化学结构差异与其抗氧化活性和生物活性之间的关系。结果表明,H型结构单元越多,木质素总酚羟基含量也越高,且木质素总酚羟基含量与其抗氧化活性(DPPH自由基清除能力和还原能力)和生物活性具有正相关关系。当木质素浓度为0.4 mg/mL时,4种不同来源木质素对DPPH自由基的清除率在14.9%~53.5%之间。4种不同来源木质素均具有提高小鼠成骨细胞活力的效果;在浓度为3.2 mg/mL时,玉米秸秆纤维素乙醇残渣木质素可使小鼠成骨细胞活力达到710%,远高于其他3种木质素。
Abstract:
      In this paper, four kinds of agricultural residue, including corn stalk cellulose ethanol residue, reed, walnut shell and rice hull, were used as raw materials to extract lignin by NaOH method. The chemical structure of lignin was analyzed by gel permeation chromatography (GPC), Fourier transform infrared spectroscopy (FT-IR) and nuclear magnetic resonance spectroscopy (NMR). The results suggested that the total phenolic hydroxyl content of lignin sample was increased with H-type structural unit content increasing. Furthermore, there was positive correlation between the total phenolic hydroxyl content of lignin with its oxidation resistance including DPPH free radical scavenging ability and reducing capacity. When the concentration of four lignin samples was 0.4 mg/mL, the corresponding DPPH free radical scavenging abilities of the lignin samples were between 14.9% and 55.5%. Interestingly, all these lignin samples could enhance the growth of mouse osteoblasts. Especially, the mouse osteoblasts cell viability of the corn stalk cellulose ethanol residue lignin could reach to 710% at a concentration of 3.2 mg/mL.
