采用Plackett-Burman设计、最陡爬坡实验、中心组合设计和响应面法分析等对实验室9种菌株处理制浆造纸废水进行最佳复合菌群构建。首先,采用Plackett-Burman设计出对制浆造纸废水降解有显著影响的4株菌种:Bacillus sp.、PE-9、Pseudomonas putida、Xz6-1。在此基础上,由最陡爬坡实验确定最大响应区域,然后通过中心组合设计和响应面法分析对显著菌株细胞浓度进行优化。优化后构建的最佳复合菌群组成及其浓度(以OD600表示)为:Bacillus sp.(0.35)、Enterobacter Cloacae(0.2)、Gordonia sp.(0.3)、Pseudomonas putida(0.38)、PB-3(0.3)、PE-9(0.41)、PG-1(0.2)、Ppp-3(0.3)、Xz6-1(0.35)。采用构建的最佳复合菌群处理制浆造纸废水,CODCr去除率为86.9%,与预测值(85.7±0.5)%接近,说明运用统计学方法构建复合菌群具有可行性。 |
Abstract: |
The microorganisms with 9 bacterial strains were constructed and optimized to enhance the treatment efficiency of pulp and paper effluent by orderly using Plackett-Burman(PB) design, steepest ascent design, central composite design and response surface method. Firstly, Plackett-Burman design was used to find the strains having significant effect on the degradation of the pulp and paper effluent, i.e. Bacillus sp., PE-9, Pseudomonas putida, Xz6-1. Secondly, the maximum response region for the central composite design was determined using the method of steepest ascent. Finally, the optimal strains concentration (OD600) was determined using central composite design and response surface method analysis. The optimized multiple microorganisms contained Bacillus sp. (0.35), Enterobacter Cloacae(0.2),Gordonia sp.(0.3),Pseudomonas putida (0.38),PB-3(0.3),PE-9(0.41),PG-1(0.2),Ppp-3(0.3),Xz6-1(0.35). Under such conditions, the CODCr degradation with multiple microorganisms was 86.9%, which was good agreement with the predicted one of (85.7±0.5)%. In conclusion, the construction of multiple microorganisms by a statistical experimental design has good reliability. |
